Testing Procedures
Easy
to perform Testing Procedures
Please note that some media and dilution materials will destroy enzymes and should not be used when testing with our FluoroQuik system. As our technology is based on the detection of enzymes produced by the target bacteria, using such materials will invalidate the test. Please inquire at FluoroVei LLC if you are unsure.
(Procedures will vary for different tests, always follow the product insert provided with each test.)
For samples not requiring any growth time, start at step #3, otherwise start at step #1.*
Please Note: If testing is performed without any growth and the result is negative, and a concern remains, a short 4 hour growth phase will allow the microorganism to begin producing enzyme, and the resulting enzyme will be easy to detect.
| *(start here if growth required) | |
| 1. Prepare the sample to be tested by adding 200 micro liters of Reagent B to 100 ml of solution sample. |

| 2. Incubate sample for 4 hours at 38.5 degrees C. |

| *(start here if no growth is required) | |
| 3. Turn on power source to the fluorometer. | ![]() |
| 4. Add 50 micro liters (2 drops) of Reagent A Substrate to the plastic test vial. | ![]() |
| 5. Add 150 micro liters of liquid sample containing potential contaminant to the plastic test vial and secure cap onto test vial. | ![]() |

|
| 6. Gently squeeze the bottom of the plastic vial two to three times to mix the solutions. | ![]() |
| 7. Holding test vial upright, shake to remove bubbles from the solution (bubbles will interfere with the reading of the fluorescence and will produce inaccurate test results). |

|
8. Place the test vial into the
fluorometer with the pointed end lined up with the indicator line near the fluorometer test chamber.

| 9. Thread the fluorometer cap onto the machine containing the sample. |

| 10. Push the Measurement button on touch screen. |
![]() |
| 11. Push button labeled “BLANK” on touch screen. (timer will start counting seconds). This function eliminates background fluorescence. |
![]() |
| 12. Push Measurement button on touch screen for initial reading. (01 will display at the top). Reading is usually 00000000. |
![]() |
| 13.Wait 5 minutes. |
![]() |
| 14.Push The Measurement button on the touch screen for the test results. (a value will be displayed in the box below the timer). Record the number from the display. Push the measure button 3 additional times to eliminate auto-fluorescent results. If the reading lowers to 00000000, the fluorescence was auto-fluorescence, and the result is negative. If the number lowers slightly, or not at all, the test is positive for the target microorganism. |
